Antharis Therapeutics is committed to accelerating immune therapies' discovery, research, development, and biomanufacturing. We are seeking a scientist who specializes in monoclonal antibodies, antibody drug conjugates and bispecific antibodies. They will be a bench scientist responsible for designing and evaluating bispecific antibody engineering strategies and novel formats. They will work collaboratively across all research functions to drive the progress of antibody technology platform development.

Job Requirements:

  • Design and construct expression vectors for antibody and protein engineering projects, including full-length monoclonal antibodies, bi-specifics, Fc-fusion proteins, and molecular glue candidates. Utilize molecular cloning, gene synthesis, and mutagenesis techniques to optimize constructs for expression and function.
  • Engineer antibody and protein variants to improve affinity, specificity, stability, or developability using rational design or library-based approaches. Examples include CDR grafting, Fc-engineering, humanization, and generation of dual-specific formats such as knobs-into-holes or scFv fusions.
  • Characterize constructs post-expression through sequencing, SDS-PAGE, Western blotting, and ELISA. Use flow cytometry for screening cell surface interactions and binding efficiency, and perform reporter assays or luciferase-based readouts to confirm mechanism-of-action for molecular glue candidates.
  • Express and purify recombinant proteins using mammalian (e.g., HEK293, CHO) and bacterial expression systems. Optimize expression conditions and scale-up production for downstream applications.
  • Conduct structure-function studies to identify key residues involved in binding or degradation. Use alanine scanning, truncation analysis, or domain-swapping to dissect mechanisms of action for engineered biologics or molecular glue degraders.
  • Implement CRISPR/Cas9 or shRNA-based methods in cell lines to assess target engagement and degradation induced by molecular glues.

· Apply statistical analysis to experimental results to ensure reproducibility, identify significant trends, and support data-driven decision-making. Use software such as GraphPad Prism, R, or Python for data visualization and quantitative analysis in internal reports, presentations, and publications.

  • Collaborate with protein sciences, screening, and in vivo pharmacology teams to test and validate constructs in both in vitro and in vivo settings.
  • Document all findings in electronic lab notebooks and contribute to technical reports, SOPs, and scientific presentations for cross-functional team meetings and regulatory filings.
  • Stay current with emerging literature and technologies in molecular biology, antibody formats, targeted protein degradation, and synthetic biology to contribute to strategic discussions.
  • Focus on achieving challenging timelines through superior planning, risk assessment, and inspiring teams to work efficiently.

Qualifications:

  • PhD in Molecular Biology, Biochemistry, Immunology, Bioengineering, or a related scientific discipline.
  • Demonstrated expertise in biologics, including mAbs, ADCs, or bi-specific antibodies.
  • Demonstrated academic or industry research experience in antibody engineering, synthetic biology, or targeted protein degradation.
  • Strong foundation in molecular cloning, protein expression systems, and data analysis tools (e.g., GraphPad Prism, R, Python).
  • Strong problem-solving skills and ability to work independently while collaborating in a team-oriented environment.
  • Excellent organizational, written, and oral communication skills.
